Extracellular Polysaccharides of Rhizobium from the Bangkok MIRCEN Collection

Thirteen of 43 strains of Rhizobium tested (8 isolated from soy bean, 2 from cowpea and 1 each from mung bean, khamin naang and puck-krached) produced large amounts of a water-soluble polysaccharide (more than 500 mg/100 ml).The components of the polysacchrides of these 13 strains were analyzed. Polysaccharides from 5 strains(TISTR 64,285,291,341 and 383; 3 isolated from soy bean and 1 each from cowpea and khamin naang) were composed of D-glucose, D-galactose and pyruvic acid in a molar ratio of 7 : 1 : 1 with some acyl groups. These were identified as succinoglycan-type polysaccharides on the basis of their components, methylation analysis and fragmentation with two specific β-glycanases of Flavobacterium. The isolate (TISTR 64) from khamin naang produced curdlan together with succinoglycantype polysaccharide. One strain (TISTR 64B) producing larfe amounts of curdlan was isolated from the stock culture. Polysaccharides from 5 isolates (TISTR 131,135,159,381 and 382; 3 isolated from soy bean and 1 each from mung bean and cowpea) contained D-glucose, D-galactose and D-mannose in a molar ration of 3 : 1 : 2,but their contents of uronic acid, pyruvic acid and acyl groups were different. A polysaccharide from 1 isolate (TISTR 139) from soy bean was composed of D-glucose, D-galactose and D-mannose in a molar ration of 5 : 1 : 2 with some acyl groups.A polysaccharide from 1 isolate (TISTR 386) from puck-krached was composed of D-glucose, D-galactose and uronic acid in a molar ratio of 3 : 1 : 1 with some pyruvic acid and acyl groups. Thus there is no clear correlation between the host specificity of organisms and the components of their extracellular polysaccharides.
