Occurrence in Alcaligenes MC11 of a NAD-dependent Dehydrogenase with High Specificity towards Ether-Alcohol Compounds

Cell-free extracts of Alcaligenes MC11 showed rather high NAD-dependent dehydrogenase activities towards some ether-alcohol compounds as well as towards ethyleneglycol and ethanol. On disc gel electrophoresis, the enzyme in extracts of cells grown on ethyleneglycol monomethyl- and monoethyl-ethers gave at least two bands (b and c), that of cells grown on diethyleneglycol gave four bands (a, b, c and d), that of cells grown on tri- and polyethyleneglycol gave two bands (a and c), that of cells grown on ethylene-glycol and ethanol gave two bands (b and c), and that of cells grown on succinic acid gave a single band (c). The enzyme in band c (enzyme c) seems to be constitutive, whereas those in the other bands (enzymes a, b and c) are inducible. Enzyme c was active with all the alcohols tested, but enzymes a b and d showed high specificity towards only ether-alcohols, although enzyme d could also act quite well on ethanol. Enzyme b was purified by ammonium sulfate fractionation and chromatography on a 10-caboxydecyl Sepharose column. It gave a single band of protein and enzyme activity on disc gel electrophoresis and had high specific activities towards di- and tri-ethyleneglycol and ethyleneglycol monoethylether, but no activity towards ethyleneglycol or ethanol.
