Isolation and Characterization of a Methanol-utilizing Bacterium Pseudomonas putida Strain 981

A new facultative methylotrophic bacterium designated as strain 981 was isolated, which grew at a high rate on methanol as the sole source of carbon and energy. The isolate was a Gram-negative, motile rod (1.0 by 2.0 to 4.0μm) with polar multitrichous flagella. This organism was identified as a strain of Pseudomonas putida (Trevisan) Migula. Cultural conditions for the growth of Pseudomonas putida strain 981 on methanol were investigated. The maximum specific growth rate was 0.45 per hr at 30℃ in a semisynthetic medium contianing 0.5% of methanol, yeast extract and simple mineral salts. The maximum growth yield was 0.5g dry cell weight per 1g of methanol.The cellular protein content and the amino acid pattern of this organism were very similar to those of the previously reported methylotroph.Biochemical characteristics of the organism are also described.
