Improvement of Stability of Fumarase Activity of Brevibacterium flavum by Immobilization with κ-Carrageenen and Polyethyleneimine

In order to improve L-malic acid production by Brevibacterium flavum immobilized with κ-carrageenan, the immobilization method was investigated. As a result, it was found that the addtion of some polycationic polymers to the immobilization medium was effective for increasing the stability of fumarase activity of the immobilized cells. Among polycationic polymers tested, polyethyleneimine was most efficient. Fumarase activity of B.flavum immobilized with 3.0% κ-carrageenan in the presence of 0.15% polyethylene-imine showed the highest heat and operational stabilities. The L-malic acid productivity of these immobilized cells increased 2 fold compared to that of B. flavum immobilized only with κ-carrageenan, and 21 fold that of B. ammoniagenes immobilized with polyacrylamide.
