Inhibition of Denitritification by Oxygen in Paracoccus denitrificans

Inhibition of denitritification (denitrification starting from nitrite) by oxygen in the anaerobically grown denitrifier Paracoccus denitrificans was examined by alternate incubation under aerobic and anaerobic conditions. Denitrification was measured by the acetylene inhibition method. The inhibiton increased with increasing oxygen concentration, whereas a high concentration of nitrite had the reverse effect. The inhibition took effect gradually with the maximum time constant of inhibition being approximately 1 h. The inhibition was quickly relieved when the cells were returned to the anaerobic condition. This recovery of denitrifying activity was due not to newly synthesized nitrite reductase (NiR), but to pre-existing enzyme. NiR remained functional ever after 22 hours' exposure to air.
