The optimal culture conditions for production of the purplish-red pigment neopurpuratin by a mixed culture of Streptomyces propurpuratus ATCC21630 and Bacillus sp. no. 751 were investigated using shake flasks and an improved basal medium.Glucose or glycerol as the carbon source and peptone and meat extract as the nitrogen sources were excellent for neopurpuratin production. The addition of ferrous sulfate to the medium was essential, and the optimal concentration relatively high (0.1-0.2%). The greatest production of neopurpuratin was observed with mixed culture in a medium consistign of 3% glucose or glycerol, 1% peptone, 1% meat extract, 0.2% FeSO_4・7H_2O, and 1% CaCO_3 (pH 7.0) at 28-30℃, under high aeration.Neopurpuratin production resulted in the best yield when Bacillus sp. no. 751 was inoculated into a comparatively early culture (e.g., an 8-h culture) of Streptomyces propurpuratus; Yield decreased when the inoculation was delayed.Under the optimal culture conditions described here, about 1,400 μg/ml of neopurpuratin was produced in 72-96 h of cultivation in a 500-ml shake flask, an improvement of 2.5 times over initial levels (500-600 μg/ml).
