Isolation and Properties of Neopurpuratin, a Purplish-red Pigment Produced by Mixed Culture

A purplish-red pigment, which we named neopurpuratin, was produced in submerged fermentation by mixed culture of Streptomyces propurpuratus with Bacillus sp. no. 751,and isolated from the broth filtrate by procedures including extraction with methanol and column chromatographies on silica gel and cellulose powder. The pigment formed very hygroscopic, deep purplish-red needle crystals and was considered to be a chelated ferrous complex. Its molecular weight was estimated to be less than 1,000 by ultrafiltration. Aqueous solution of the pigment is vivid purplish-red and shows uv absorption maxima at 278 nm (E^<1%>_<1cm> 387.5) and 558 nm (E^<1%>_<1cm> 167.5). This pigment has potential use as a natural food coloring agent because of its water-solubility and negligible toxicity in biological tests.
