Mixed culture interactions between R. capsulata and a Klebsiella sp. growing on defined glucose media in batch and continuous culture have been studied in order to understand the role of photosynthetic bacteria in the treatment of high BOD wastes. Sequential conversion of glucose to volatile fatty acids (mainly acetic) by the Klebsiella sp. occured, followed by their utilization by R. capsulata. The autolysis of cells of Klebsiella sp. also stimulated the growth photosynthetic bacteria. Light-energy limitation effects were studied. The dissolved oxygen tension (DOT) was found to be a critical environmental parameter with partially aerobic conditions (DOT=1% air saturation) being optimal for growth of R. capsulata and for COD reduction. In single-stage continuous culture, maximal growth of R. capsulata occured at low dilution rates (D less than 0.10 hr<-1>). Two-stage continuous culture with a longer residence time in the second stage was found to be advantageous in effecting increased assimilation of volatile fatty acids.
