Histopathological Reaction of a Calcium Phosphate Cement Root Canal Filler

A calcium phosphate cement (CPC) paste, consisting of Ca_4 (PO_4)_4 O and CaHPO_4 mixed witheither glycerol or polyethylene glycol, was shown to provide good sealing ability when used as a root canal sealer/filler. The present study compared the in vivo aspects of CPC paste with two commercially available endodontic materials, Grossman's cement (GC) and a calcium hydroxide-iodine paste (Vitapex), using a dog model. Histopathological reactions of the materials were evaluated by filling the materials in root canals prepared accordig to standard endodontic procedures. The dogs were sacrificed at 2 weeks to 6 months after the operation, and the periapical tissues including the root apex were excised. Histological finding obtained from the CPC group showed that slight inflammatory reactions occurred in the periapical tissues. Newly formed cementum and bone tissues were also observed. Severe inflammation was provoked by GC, and absorption of roots and bone tissues in periapical tissues was still obsevable at 6 months after the operation. Tissue reactions of Vitapex in the short-term period(< 1 month) were similar to those of GC. Six months after the operation, slight inflammatory reactions were still observed in the periapical tissues. The results showed that CPC is highly compatible to periapical tissue, and point to the possibility that CPC may be used in simplified endodontic procedures.
