The experiments, whose materials were all leaved stems of Portulaca grandiflora, were pursuited under following procedures. 1. Of leaved stem cuttings with unexcised apices. After a 3 or 6 hours soak in 1/2×lO^<-4>NAA solution (as solvent distiled water being employed), the cuttings were cultured respectively in distiled water, pure city water and Kasugai's solution. 2. Of leaved stem cuttings with excised apices. They were cultured in pure city.water, after a 24 hours soak in pure city water or 1/2×lO^<-4>NAA solution (as solvent distiled water being employed). 3. Of leaved stem cuttings with unexcised or excised apices. Simultaneous comparison of the above both kinds of materials by culturing in pure city water, after a 24 hours soak in pure city water or 1/2×lO^<-4>NAA solution. 4. Of leaved stem cuttings with unexcised or excised apices. Similarly, they were cultured in pure city water, after a 24 hours soak in pure city water or 1/2×lO^<-4>NAA solution. But in this case, one half of materials were kept in dark during soak period, while the other in light throughout the experiment. As above descrived, the experiments were carried cut urcer such differer.t conditions, therefore the results obtained amounted to a great deal, of which only the most significant facts were briefly summarised below. 1. When cultured in Kasugai's solution after a soak treatment with NAA solution, the responses of cuttings were usually greatest, as compared with the cases, when cultured in water only or cultured in Kasugai's solution without such a soak treatment. 2. When cuttings were excised, the effects of NAA soak on the root responses were most obvious. 3. As often dealt with by the writer, the cuttings, when screened with fit intensity, brought better effects on root responses than unscreened. 4. After all, from the facts above mentioned, it should be able to conclude that when these four factors i.e. excision, soak, screening and culture solution were combined with best conditions, the regenerating results came to maximum.
