Active and inactive renin concentrations and their molecular weights in plasma and the kidney were investigated in relation to the elevations of blood pressure in stroke-prone spontaneously hypertensive rats (SHRSP) and malignant or precocious SHRSP (M-SHRSP). Active and inactive plasma renin concentrations (PRC) in 6-week-old M-SHRSP were almost the same as those in WKY. In 12-week-old rats with the blood pressure over 250±6.3 mmHg, however, both active and inactive PRC increased. Although active renal renin concentration (RRC) values in 6-week-old M-SHRSP were almost the same as those in WKY, inactive RRC increased to approximately ten times that of WKY. Both active and inactive RRC increased about 1.5-fold at the age of 12 weeks. Plasma renin molecular weight in WKY and hypertensive rats with the blood pressure under 200 mmHg was 38,000 (low molecular weight renin : LMR), but the high molecular weight renin (HMR) was also recognized after the blood pressure elevation above 200 mmHg. Following captopril treatment, PRC in WKY, SHR and 7-week-old M-SHRSP increased significantly, in contrast to a remarkable decrease in 10-week-old M-SHRSP. At the same time, HMR concentration decreased and LMR concentration increased. The average life spans of the rat treated with captopril increased nearly 1.5-fold. For 12-week-old M-SHRSP, however, PRC levels decreased, and neither transition from HMR to LMR nor lengthening in the life span were observed. These results indicate the qualitative and the quantitative changes in renin are related to the elevations in blood pressure in the SHRSP and M-SHRSP with severe hypertension.
