セロビオヒドロラーゼの反応速度論的解析 : 固液界面における酵素反応を表面密度という概念を用いて定量化する

自然界において結晶性セルロース大部分は,種々の微生物が生産するセロビオヒドロラーゼ(CBH)によって分解されている。しかしながら,CBH の反応には基質,酵素双方の性質が関与しており,さらに反応が固液界面で進むという特徴を有していることから,定量的な解析が非常に難しい。そこで本研究では,結晶性セルロースの表面積を CBH の最大吸着量(Amax)から見積もり,反応中の吸着量(A)を Amax で割った表面密度(ρ=A/Amax)に対して,吸着した酵素一分子あたりの速度(k=v/A)をプロットすることで基質の表面積を相殺した。その結果,基質の性質がセロビオヒドロラーゼの反応速度に大きく影響していることが示唆された。
