1. The effect as well as the site of action of N-(diethylaminoethyl)-2-methoxy-4-amino-5-chlorobenzamide dihydrochloride (métoclopramide) upon the motility of the isolated small intestine of various mammals, especially of rats were studied.<BR>2.(a) The drug usually produced an augmentation of the intestinal motility.(b) After applying the ganglion blocking agents such as nicotine or hexamethonium as well as atropine, the drug produced an excitatory effect not inferior to that observed before the application of the agents described above. On the other hand, (c) after applying the drug to the intestine, the ganglion stimulating agent, DMPP, became ineffective and, moreover, the excitatory action of nicotine could not be observed. And (d) soon after the application of the large dose of the drug the stimulation of the mesenteric nerves proved to be ineffective.<BR>3. After the intestinal tone was extremely raised by applying the large dose of the drug to the intestine, further application of the drug produced a remarkable decrease of the tone.<BR>4. From the results described above it may be concluded that (a) métoclopramide exerts a paralytic action on the intramural nervous elements, (b) while it exerts an excitatory action on the intestinal muscle, which can, however, be inhibited under some circumstances, e.g., when the tone is extremely high.
