Studies of cell cultures in human spinal ligaments.

In order to make clear the mechanism of spinal ligament ossification, we used cultured cells, obtained by enzymatic means from human spinal ligaments. After release from the tissue, cells were cultured in dishes in DMEM+Fetal Bovine Serum. After two or three days, microscopically visible colonies appeared at plating efficiencies of 5-10% of the starting cell numbers. Morphologically they varied from roughly polygonal cells to typically spindle-shaped cells. Cultured cells were divided into two groups. Cells which were obtained from non-ossified ligaments from patients with ossified spinal ligaments, were put into group A. Cells, obtained from patients with non-ossified spinal ligaments, were put into group B. We made a comparative study of them. Cultured cells in group A showed more numerous dendrites and wide cytoplasmic spaces, suggesting high cell activity. In the cAMP production, group A showed a positive response to PTH. PTH suppressed alkaline phosphatase activity in group A. These changes seem to suggest that there might be a possible difference in the charactaristics of two groups.
