The endocochlear potential (EP) was recorded by a micropipette filled with 3 molar KCl solution, which was introduced through the spiral ligament into scala media. The average magnitude of EP in the basal turn was 100mV. and in the fourth turn approximately 50mV, and the potential gradient was always found along the cochlear duct.EP was sensitive to the oxygen lack and the reversal of its polarity was observed in the anoxic period. Ep showed complete recovery within few minutes after the anoxic anoxia lasting 5 minutes. When the curarized guinea pig was killed by cutting the oxygen supply off, the negative EP reached to about 50% of the initial EP value and the gradually reached to zero about 2 hours after death, while the negative EP in the histotoxic anoxia induced by applying KCN solution to the cochlea disappeared in shorter period than 2 hours. The negative DC potential inside the organ of Corti was still present when EP changed its polarity. Cochlear microphonics was more sensitive to anoxia than EP.The change of EP in response to acoustic stimuli was nothing but summating potential (SP). The magnitude of SP in scala media was much larger than in the perilymphatic space and the phase in scala media is opposite to that in scala tympani. The magnitude or the phase of SP in scala media is a function of the distance between the measuring point and the round window. It was revealed that the source of SP is located on the hair bearing end of the hair cells and the DC shift in response to tonal stimulation was added to the EP so that it seems to be a change of EP.The hyperpolarization of the cuticular layer increased EP and the negative component of SP, while its depolarization decreased both EP and SP. This result was coincide with the observation of the effect of DC current across the cuticular layer on EP and SP. Therefore SP may be generated in the hair cells, the polarity of which may depend on the resultant of unsymmetrical vibration of the basilar membrane.
