A study of the relationship between nuclear DNA content and pathologic findings and prognosis in colorectal cancer.

The nuclear DNA content was measured by microspectrophotometry in 70 patients in order to evaluate the malignancy of individual colorectal cancers.<BR>For the nuclear DNA analysis, specimens were obtained by biopsy or sugery and DNA distribution patterns were divided into types I, II or III according to the degree of dispersion on the DNA histogram.<BR>The DNA histogram pattern was not correlated with the location and length of the tumor, depth of invasion, lymph vessel invasion or lymph node metastasis. In cases of venous invasion or liver metastasis, the type III histogram was predominant, whereas in cases without these features, the type I histogram was predominant. Type I was observed significantly more often among patients who survived for more than five years than in those who died within five years. Type I cancers showed a better prognosis, but in the more advanced group, the risk of recurrence appeared to increase even in such cancers.<BR>These results suggest that the DNA content of the cancer cell nucleus may be one of the most important prognostic factors in colorectal cancers. Thus, data obtained by DNA analysis of biopsied specimens seems to be useful for providing information on the prognosis of colorectal cancers before surgery.
