Herbaceous cuttings of Rehmannia glutinosa Lib. var. purprea MAKINO obtained from in vitro culture were planted in vermiculite. The optimal temperature for growth was determined by using Biotron. As regards rooting rate, root number and root length, the most favourable temperature for rooting was 25℃. Root formation was slower at 20℃ than at 25℃. All the cuttings died within 10 days when the temperature was kept at 30℃. Leaves cultured in vitro on MS medium containing IAA formed roots within a week; addition of 1 ppm IAA caused root formation in 64% of the leaf explants. When the rooted leaf segments were subcultured on a medium containing 2.5 ppm BAP, multiple shoot formation occurred within 5 weeks. In order to get direct shoot formation from leaf segments, leaf cuttings were cultured on MS medium containing BAP or both BAP and GA. Although numerous shoot primordia were formed on the surface within 5 weeks, their growth was very poor. On the other hand, all the leaf cuttings in soil died within a week. Instead of storing roots before planting, plants obtained from the cuttings were immediately cultivated at 25℃ with watering once in 7-10 days for one year. These plants grew vigorously when cultivated in the greenhouse.
