Callus was induced from stem segments of Scopolia (collected in Miyazaki) on Murashige and Skoog (MS) medium containing 2,4-D and kinetin (0.25-0.25 mg/l) under 24 hr light exposure at 25°. Differentiation into shoot, root and occasional plantlet was observed during the callus multiplication. Shoot differentiation was also observed in callus culture. Seventy-eight% of shoot cuttings formed root on NAA (0.5 mg/l)-containing MS medium, and 53% of them were successfully transplanted to soil. In the case of Scopolia collected in Shikoku, Honshu and Korea, callus was induced from stem segments on MS medium containing 2,4-D and kinetin (0.25-0.25 mg/l). However, shoot dif- ferentiation from those callus was seldom observed. Shoot cuttings from shoot-mass from the callus of Miyazaki Scopolia formed root on MS medium containing IAA (1 mg/l): the plantlets were successfully transplanted to sterilized vermiculite.
