Pharmacological Studies on Iridoid Compounds. I : General Pharmacological Action of Patrinoside and Its Aglycone

General screening tests were made on patrinoside, which is commonly contained in the plants of Valerianaceae, and on its aglycone. LD_<50> values of patrinoside in mice were 595 mg/kg (i. v.), 350 mg/kg (s. c.), 555 mg/kg (i. p.), and > 2g/kg (p. o.), and those of its aglycone were 143 mg/kg (i. v.), 234 mg/kg (s. c), 182 mg/kg (i. p.), and > 500 mg/kg (p. o.). Both patrinoside and its aglycone showed hypothermia at doses higher than 1/5-1/4 of LD_<50>, and patrinoside inhibited the writhing movement induced by acetic acid in mice. Both patrinoside and its aglycone lowered blood pressure and heart rate in rats and rabbits. Patrinoside aglycone slightly inhibited acetylcholine-induced contraction of isolated mouse ileum and histamine-induced contraction of the isolated guinea pig ileum at high concentrations. However patrinoside itself did not inhibit their contraction. Patrinoside induced cathartic action when administered orally while patrinoside aglycone failed to it. Bile secretion was increased markedly by patrinoside administered intravenously or intraduodenaly and onlly slightly by its aglycone.
