Since Edkins many investigators have studied the problem of the role of the pylorus in gastric secretion, but the results show many discrepancies and the mechanism is not understood in either the gastric phase or the intestinal phase of gastric secretion. In these experiments the role of the pylorus was studied during both the gastric and the intestinal phases of gastric secretion in an attempt to arrive at a definite conclusion. For this purpose two types of whole stomach pouch dog were used. The one had a whole stomach pouch with duodeno-esophageal anastomosis with sectioned vagi, and the other was the same except that the pyloric portion of the stomach had been entirely removed. Liver extract No. 343 (Lilly) was used as the experimental substance. 4.2gm. of liver extract was dissolved in 50cc. of water and applied to the gastric mucosa during 30 minutes and 60 minutes respectively on both animals to investigate the role of the pylorus during the gastric phase of gastric secretion; and liver extract (4.2gm.) dissolved in 30cc. of water was given by mouth to both animals to show the role of the pylorus during the intestinal phase of gastric secretion. In the latter experiment, of course, the secretagogues entered the intestine directly. All these experiments were performed when the intestinal phase of gastric secretion was absent. These experiments warrant the following conclusion: The pyloric portion of the stomach plays an important role in gastric secretion due to chemical stimulation during the gastric phase, but it does not exert any important chemical stimulation during the intestinal phase.
