Numerous investigators have administered various known secretagogues parenterally in order to determine whether secretagogues stimulate gastric secretion after absorption into the blood stream. However, such substances have not been shown to be free from vasodepressor substances such as histamine or toxic substances. Thus the physiological interpretation of the results observed is doubtful. In my studies certain known secretagogue preparations such as liver extract Lilly No. 343 and Liebig's meat extract were administered parenterally both in the original form and after rendering them free from vasodepressor substances. By using both whole stomach pouch and Pavlov dogs, I tried to establish the effectiveness of the secretagogues present in food both before or after digestion, after absorption into the blood stream. All these experiments were performed on animals not at the intestinal phase of gastric secretion in the case of whole stomach pouch dogs, and free from both the gastric and the intestinal phases in the case of Pavlov pouch dogs. The results may be summarized as follows: 1) Intravenous injection of vasodepressor-containing liver extract was more effective in promoting gastric secretion than vasodepressor-free extract. But vasodepressor-free liver extract does stimulate gastric secretion after intravenous injection. This shows that nontoxic secretagogues are able to stimulate gastric secretion after being absorbed. 2) The results of parenteral administration of vasodepressor-free secretagogues by various routes are not the same. This indicates that the rates of absorption are different, for as stimulation by secretagogues injected intravenously always occurs at a constant rate, where as by other routes the results are not constant, we may assume that accumulation in the blood stream after injection is necessary. 3) Atropine prevents the action of vasodepressor-free secretagogues injected intravenously.
