Isolation and characterization of three protease-producing bacteria from a deep-sea mud of Japan Sea

We have isolated three protease-producing bacteria from a deep-sea mud of Japan Sea, by observation of the growth with disappearance of casein-color (halo generation) on skim-milk containing marine agar medium. It is suggested that these bacteria designated as strains E1, G1 and A5, were mesophilic isolates, for these optimal growth at 37C. Relative protease activities of these isolates were, however, varied between strains from at low (8C) to high temperature (50C). In addition, one of selected bacterium, strain E1, was characterized in some aspects. Growth curves in the strain E1 were observed at 20, 30, 37 and 45C, respectively. The calculation by logarithmic slope of the growth at each temperature, showed the generation time of 4.6h at 20C, 1.7h at 20C, 1.25h at 20C, 3.5h at 20C, respectively. Light microscopic observation revealed that isolated bacterium, strain E1 is classified to "coccus" morphology.
