Two plasmids derived from halophile-specific plasmid pWL102 with upstream and downstream region of rrn operons, were constructed, in order to disrupt rrn operons, rrnA and rrnB on chromosomes in extremely halophilic archaeon Haloarcula japonica strain TR-1. One of these plasmids, pDT2, for disruption of rrnA operon, is derived from DNA fragments of upstream (676 bp) downstream region (1, 072 bp) linked with the 27 bp of synthetic linker. Second plasmid, pDT3, for disruption of rrnB operon, is also derived from DNA fragments of upstream (668 bp) downstream region (597 bp) linked with the 27 by of synthetic linker. Gene disruption of rrn operons on the chromosomes in extremely halophilic archaeon H. japonica strain TR-1, using these plasmids with homologous recombination, is definitely not only essential, but also useful, to obtain mutants disrupted in the genes of rrn operons.
