Epidermal growth factor induced apoptosis in the outer root sheath cells of anagen hair and changed the hair cycle to the catagen phase

Objectives : The effects of epidermal growth factor (EGF) on individual hairs are different depending on the phase of the hair cycle. We investigated the effect on anagen hair and the mechanism of hair loss with EGF administration using sheep as a model of an anagen synchronized animal. Materials and Methods : Systemic hair loss was induced in a short period of time following a single subcutaneous administration of EGF at a high dose to a sheep. Skin biopsy was performed and serum samples were collected according to a schedule, followed by gathering of various data. Histopathological changes, h-rEGF concentrations in the blood, h-rEGF localization sites, and apoptotic cell localizations were observed following administration of h-rEGF. Results : Acute hair loss was observed after a large dose of h-rEGF. At 6 hours after administration, the serum h-rEGF level decreased to half of its original value and numerous h-rEGF localizations were observed midway on the hair strand. Outer root sheath cells became apoptotic during the same period, and the anagen phase changed into catagen. Hair loss occurred 6 days after administration. Conclusion : EGF induced apoptosis in anagen hair and changed the hair cycle to catagen.
