Synthesis and Separation of 18β-Glycyrrhetyl Monoglucuronide from Serum of a Patient with Glycyrrhizin-Induced Pseudo-aldosteronism(Organic,Chemical)

18β-Glycyrrhet-3-yl- and 18β-glycyrrhet-30-yl-β-D-glucopyranuronic acid (3MGA and 30MGA) were prepared from 18β-glycyrrhetic acid (GA) by the Koenigs-Knorr synthesis, and their retention times on high-performance liquid chromatography (HPLC) were compared with that of GA-conjugate which was separated from the serum of a patient with pseudo-aldosteronism who had received large doses of 18β-glycyrrhizin (GL) for the treatment of liver disease. The results indicated that GA-conjugate was identical with synthetic 3MGA. The hydrolysis of GA-conjugate and 3MGA with glycyrrhizin hydrolase(β-glucuronidase from Aspergillus niger GRM3) gave glycyrrhetic acid (GA), but 30MGA was not hydrolyzed by the same enzyme. The concentrations of 3MGA, GL and GA in the serum of this patient were measured by HPLC.
