Experimental Studies on the Adrenal Cortex of the Mouse-Fetus : II. Effects of Various Treatments of the Mothers on the Adrenal of the Fetus

(1) Hypophysectomy of the mother resulted in an atrophy of the maternal adrenal, but caused almost no change in the size and histological configuration of the fetal adrenal. (2) Administration of ACTH at daily doses over 1 i.u. to the mother resulted in an enlargement of the fetal as well as the maternal adrenal. The lipid droplets included in the cortical cells of the fetal adrenal tended to be smaller in size. Administration of ACTH at daily doses of 0.3 and 0.5 i.u. caused also an enlargement of the maternal adrenal, but produced almost no change in the fetal adrenal. (3) Injection of cortisone to the mother, either with or without maternal adrenalectomy, retarded the growth of the fetal adrenal, reduced the lipid content in the gland and enlarged the size of the lipid granules. (4) Short-term injection of formrlin increased the weight of the maternal adrenal, but decreased the size of the fetal adrenal, in which the lipid granules tended to be enlarged. Prolonged injection of higher level of formalin produced the increase of the maternal adrenal which became hyperaemic and flaccid in appearance, and gave slight effects on the fetal adrenal. (5) These results suggest that large amount of exogenous ACTH passes through the placenta and stimulates the fetal adrenal, while a moderate amount does not stimulate the fetal adrenal in spite of having capacity for enlarging the maternal adrenal. It was not demonstrated that endogenous ACTH passed through the placenta and showed stimulating effects on the fetal adrenal.
