Modulation of Neutrophil Apoptosis in Rheumatoid Arthritis

Infiltration of large numbers of polymorphonuclear cells (PMN) into synovial fluids is characteristic in rheumatoid arthritis (RA). PMN have been shown to play an important role in inflammation and tissue damage in RA. In order to evaluate the fate of infiltrated PMN, the role of synovial fluids (SF) and conditioned medium of rheumatoid synovial cells (SC) on spontaneous apoptosis of cultured PMN was investigated. Apoptosis of PMN was determined by morphological changes, flow cytometrical analysis, and DNA fragmentation. SF from patients with RA and the conditioned medium of RA-SC significantly decreased the rate of spontaneous apoptosis of cultured PMN. These effects were partially alleviated by the treatment with neutralizing antibody for granulocyte-macrophage colony stimulating factor (GM-CSF). The results indicate that the retardation of PMN apoptosis may contribute to the perpetuation of synovitis and the enhancement of joint damage in RA.
