Establishment of a New Tumor Marker : Sera from Cancer Patients Induce Lower Activities of Phosphofructokinase (PFK)

A new tumor marker test, the "PFK-Assay Method", has been developed based on the finding that sera derived from cancer patients induce lower activities of "phosphofructokinase" (D-fructose-6-phosphate 1-phosphotransferase, EC 2. 7. 1. 11) (PFK) after having examined every condition in the reaction, such as incubation time for every reaction component, and amounts of enzymes and substrates. The most unique aspect of the new PFK assay method is the application of mixing method of an ATP-aqueous solution with a low pH of 3.2 into the mixture of PFK and serum prior to injecting the main reaction cocktail. It was impossible to distinguish sera of cancer patients from those of normal healthy individuals when ATP was mixed into the reaction cocktail consisting of Tris buffer pH 8.5, or when the ATP solution was adjusted to a neutral range. Clinical application of the PFK-assay method revealed the effectiveness of the method in distinguishing cancer patients from normal healthy individuals. Sera from cancer patients tended to show a lower buffering capacity against acidic ATP solution, and this might be one of the causative factors which produce differences between sera from cancer patients and those of normal healthy individuals.
