Changes in Enzyme Activities Recognized in Lymphocytes from Patients with Carcinomas of the Gastrointestinal Tract

Changes in the activities of ATPase and lysosomal enzymes in lymphocytes from patients with carcinomas of the gastrointestinal tract (GI carcinoma) in different performance status and different immunological status were investigated in order to evaluate whether or not the changes in these enzyme activities correlate to the immune responsiveness of carcinoma-bearing patients. Three hundred patients with GI carcinoma were selected as subjects and 150 normal individuals as well as 90 patients with nonmalignant GI disease were also selected as controls. The criteria of Karnofsky's performance status were used as performance status. Mononuclear cells were separated on the standard Ficoll-Conray density gradient. After preparing the crude enzyme, the activities of ATPase, acid phosphatase and β-glucuronidase were determined by a modification of Skou's method, a modification of Bessey's method and our devised modification of P-nitrophenyl β-glucuronide-using method, respectively. The experimental data indicate that the changes in the activities of enzymes such as oligomycin-sensitive ATPase, bound form of acid phosphatase and β-glucuronidase in lymphocytes correlated to the immunological parameters in patients with GI carcinoma. Therefore these enzyme activities in lymphocytes may be evaluated as the available parameters which reflect the immune responsiveness in patients with GI carcinoma, though the changes in these enzyme activities are not considered to be specific in patients with GI carcinoma.
