Effects of cytochalasin D and colchicine on the mechanically stretched periodontal ligament fibroblasts in vitro and in vivo

The tension remaining within the periodontal ligament (PDL) is regarded as a part of the causes of relapse in and out of the retention period after orthodontic tooth movement. In this study, we investigated the relationship between the cytoskeleton of PDL fibroblasts and their contractile activity in order to unravel the intracellular mechanisms of relapse. We employed the experimental model of multilayered human PDL fibroblasts cultured between human dental root discs in vitro. When cytochalasin D, an inhibitor for actin polymerization was applied to this system, a greater extension of inter-disc space (IDS) was observed in response to the stretching force application compared to the control group without cytochalasin D treatment. Similar but slightly weaker effects were also observed by the treatment of colchicine, an inhibitor for tubulin polymerization. The recovery of IDS after the stretching was inhibited by cytochalasin D, yet the inhibitory effects of colchicine were not as obvious as those of cytochalasin D. Furthermore, injection of cytochalasin D into rat gingival tissues in vivo prior to the separation of the maxillary incisors dose-dependently reduced the recovery movement of the incisor after the removal of the separating elastic. These results suggest that the actin filaments may be closely related with the contractile activity of PDL fibroblasts and may play important roles in the relapse after orthodontic tooth movement.
