An improved technique for separation of glandular trichomes from leaves of rugosa rose (Rosa rugosa Thunb.) was developed. The glandular hairs that had been tightly attached to the tissue of leaf veins via rigid cells were efficiently detached by shaking with quartz sand dispersed in a solution of mannitol and sodium ascorbate. The mixture of the glandular and simple trichomes suspended in the separation solution was collected by filtration over nylon mesh (150 mesh), and then it was centrifuged in a solution that contained glycerol. The glandular trichome tips, which were not susceptible to browning, were used for preliminary studies of cell-free conversions of sesquiterpenes. The glandular trichomes were found to be a potential source of enzymes associated with the biosynthesis and/or bioconversion of sesquiterpenes of R. rugosa.
