Betacyanin-Decolorizing Enzymes from Phytolacca americana

Betacyanin-decolorizing enzymes (BDEs) in Phytolacca americana were partially purified and characterized. Acidic enzymes, BDE_1 and BDE_2, and basic enzymes, BDE_3 and BDE_4, were partially purified by column chromatography on DEAE-Sepharose CL-6B and CM-Sepharose CL-6B. These enzymes degraded betanin to its aglycon (betanidin) and then to a product with a peak of absorbance at 485 nm. The optimum pH values were 5.0 for BDE_1, 3.5 for BDE_2, and 5.5 for BDE_3 and BDE_4. BDE activity was detected in extracts of the leaf, stem, seed, and root. The highest specific activity of BDE_2 was found in the red epidermis of the stem. The activity of BDE_2 was inhibited by NH_4NO_3, KNO_3, KCl, and NaCl, but the activities of the other BDEs were not inhibited. These results suggest that BDEs Other than BDE_2 control the degradation of betacyanin in the intracellular space.
