Lipid Biosynthesis in Epidermal, Guard and Mesophyll Cell Protoplasts from Leaves of Vicia faba L.

Protoplasts of epidermal cells, guard cells and mesophyll cells (ECPs, GCPs and MCPs) simultaneously were prepared enzymatically from light-grown leaves of Vicia faba then purified by discontinuous density gradient centrifugation in Percoll. Although the ECPs contained small chlorophyllous plastids, they showed no measurable photosynthetic carbon fixation activity. Comparable rates of the incorporation of [^<14>C]acetate into the lipid fraction per cell was obtained for ECP, GCP and MCP. Distribution of radioactivity in the lipid classes and fatty acid species was similar in GCP and ECP but differed markedly in MCP. The ability to synthesize C_<20> and C_<22> fatty acids was present in GCP and ECP but not in MCP.
