Development of RNase activity in embryonic axes of germinating pea seeds

RNase activity in embryonic pea axes increased in parallel with the rise of RNA synthesis as germination proceeded. The development of this enzymatic activity was modified antagonistically by application of GA_3 and ABA and inhibited severely by treatment with CH. Sedimentation analysis of ^3H-adenosine-labeled RNA indicated that the synthesis of all types of RNA species is uniformly stimulated by GA_3 and inhibited by ABA. However, 5-FU treatments, which severely inhibited the synthesis of rRNA, with a slight effect on that of mRNA, had no appreciable effect on the development of RNase activity in the axes. These results indicate that active RNA synthesis during germination is independent of the development of RNase activity and that the de novo synthesis of RNases may be controlled by the synthesis of their specific mRNAs. Among the three types of RNase (RNase I, II and III) detected in the embryonic axes, RNase III showed a sharp increase in activity with embryo growth and the activity of this enzyme was mainly associated with the endoplasmic reticulum.
