Frozen and thawed cells, as well as sonicated cell preparations of Chlorella protothecoides, were assayed for activity to synthesize porphyrins from added ALA or PBG. Activity was very low in etiolated cells, and markedly developed during the process of light-induced greening. The development of activity was strongly suppressed by glucose. Activity for the synthesis of URO(gen) from ALA was initially developed, then the formation of COPRO (gen)-synthesizing activity ensued. In glucose-suppressed cells as in cells incubated in continuous darkness, URO was the main porphyrin produced and COPRO was virtually missing in the reaction products with added ALA, indicating that development of activity for the conversion of URO(gen) to COPRO(gen) is greatly enhanced by light and is repressed by added glucose. Suppressive effects of CP and CH on the development of porphyrin-synthesizing activity were also studied. From these and other results, a tentative scheme is presented for the enhancing effects of light and the suppressive effects of glucose on the development of porphyrin-synthesizing activity in etiolated algal cells, in correlation with the effects on other processes of chlorophyll biosynthesis.
