Localization of type XV and type XVIII collagens in blood capillary basement membrane

Aim: Type XV and type XVIII collagens comprise endostatin and restin in their C-terminal parts, respectively, which have been shown to have anti-angiogenic activity. Both collagens localize mainly in basement membrane (BM), but their distributions in blood vessels are not fully studied yet. Method: Localization of the both collagens in the blood vessels was studied using indirect immunofluorescence and double immunostaining. Monoclonal antibodies used were CH15A specific for human al(XV) and CH18B and CH18D for al(XVIII) chains. Results: Subendothelial BM of the capillaries in skin epidermis, renal interstitiutn, and mucous of intestines contained both collagens in general. But specialized capillaries, such as glomerular and alveolar capillaries, liver sinusoid, and splenic sinus expressed only al(XVIII) chain. On the other hand, those among cardiac, skeletal and smooth muscles exhibit al (XV) chain predominantly. Conclusions: The variety of the expression of both collagens was demonstrated in the capillary BMs. This result implies that the expression difference may depend on the function and/or structure of the blood capillaries.
