CYTOPHOTOMETRIC DETERMINATION OF GLUTAMIC DEHYDROGENASE ACTIVITY IN MOUSE BRAIN BY USING TRIDENSITY-AUTOMICROPHOTOMETER

The TRIDENT, a specially devised microphotometer for dehydrogenase assay in tissue sections, was applied to examine the distribution of glutamic dehydrogenase (GDH) activity in tissue sections of the mouse brain. With this system, the activity, relative activity (RA), was represented by the color intensity of formazan due to enzymatic activity per tissue protein. The RA value obtained increased linearly with the incubation time up to 20 min in various regions of the mouse brain. The GDH activity was quantitatively compared between four regions in the cerebellum, the cerebral cortex, hippocampus, substantia nigra and putamen in determining the RA per min. The cerebral cortex and hippocampus were relatively high in GDH activity. In the cerebellum, GDH activity of the granular layer was the highest of the four regions. Thus, the TRIDENT was highly useful in quantitative examination of the discrete distribution of the enzyme activity of tissue sections.
