The separation of mycelia of Streptomyces griseus from the cultured broth by filtration requires a considerable length of time dur to a marked resistance of the cake against liquid flow in filtration.The network of fibrous mycelia and non-Newtonian characteristics of the broth are considered to be mainly responsilbe for it.In this study, a seried of experiments was made to find ways of accelerating the velocity of this type of filtration. If the temperature of the broth had been raised to 80-90℃ and the pH adjusted to 3.5-4.0 and in addition, 2%(wt/vol) diatomaceous earth(filter aid) had been added prior to filtration, then the cake showed a conspicuous decrease of specific resistance, to an extent of 1/100,as compared with the usual practice.The apparent increase of effective pore size within the cake as well as the decrease of liquid viscosity owing to the coagulation of mycelial protein in the pretreatment, could account for the satisfactory operation. The recovery yield of streptomycin M with this procedure of filtration was found to be excellent.
