Previously I reported about the improved method of one-dimensional diffusion method in amylase assay. This report deals with the influence of the following factors on the estimated values of diastase, pancreatin and Taka-diastase : starch and agar concentrations, pH of agar-starch column, pH of amylase solution which is laid on the agar-starch column, and temperature. The conclusions are as follows. 1) In the case of constant concentration of amylase solution, square of the estimated value (the length of decoloured agar-starch column) was proportional to the logarithm of starch concentration when agar concentration was constant, and was proportional to the logarithm of agar concentration when starch concentration was constant. 2) The estimated value was affected by pH of agar-starch column and each amylase was found to diffuse best at the following pH. [table] But the minimum available concentration of amylase remained constant even when the pH of agar-starch column was varied. 3) The estimated value was affected by pH of amylase solution and the best diffusion of each amylase occurred at the same pH as the above mentioned. The minimum available concentration of amylase was also affected by pH of amylase solution and the minimum value of each amylase was obtained at the following pH. [table] 4) Both the estimated value and the minimum available concentration of amylase were affected by temperature and below 50℃ the estimated value was proportional to the temperature.
