Optimal conditions for protoplast formation and regeneration in Bifidobacterium pseudolongum and Bifidobacterium longum were determined. Protoplasts were formed by lysozyme treatment together with protease or crude α-amylase showing proteolytic activity. Cell wall digestion of both strains was insufficient when the cells were treated with lysozyme alone. Regeneration of protoplasts from both strains was acomplished with a hypertonic medium containing raffinose, MgCl_2 and CaCl_2 as stabilizers. Using optimal conditions, it was possible to prepare protoplasts of both strains and regenerate them with a frequency of 10^<-3> to 10^<-1>.
