The production of 5'-guanylic acid (GMP) by enzymatic conversion of 5'-xanthylic acid (XMP) was investigated. Resting cells of Brevibacterium ammoniagenes KY13510,which are prectically devoid of 5'-nucleotide-degrading activity and have an elevated level of XMP aminase, were used as the enzyme source.A surfactant, polyoxyethylene stearylamine (POESA), which enabled XMP and GMP to permeate the cell membrane, was required for the conversion. Phosphate ion, magnesium ion, and glucose were also shown to be essential for the conversion.Significant amounts of GDP, GTP, and guanine, as well as GMP, were accumulated at 32℃; but at 42℃ more than eighty percent of XMP consumed during the conversion reaction was converted to GMP. Aeration was indispensable, and optimal aeration and agitation were respectively one vvm and 450 rpm in a 5-l jar fermentor. pH, controlled with ammonia solution, was optimal at 7.4.From approximately 50 mg of XMPNa_2・7H_2O per ml, 43.5 mg of GMP・Na_2・7H_2O per ml was poroduced in 22 h. The mechanism of ATP regeneration from ADP, phosphate ion, and glucose, which is requisite for the conversion reaction, is discussed.
