Production of ATP from Adenine by Brevibacterium ammoniagenes

In order to develop a process fot the production of ATP from adenine with Mn^<2+>-unlimited cells, instead of previously reported Mn^<2+>-limited cells^<1,2)> of Brevibacterium ammoniagenes, the screening of agents which might allow ATP to permeate through the cell membrane and accumulate in the reaction mixture was carried out. Among 17 agents tested, 4 surfactants and 3 organic solvents were effective. Polyoxyethylene stearylamine, a cationic detergent, was the most effective and was used in the rest of this study.With resting cells of B. ammoniagenes KY13510 grown in Mn^<2+> rich medium, the reaction conditions for ATP production from adenine were optimized at 33℃ and a pH of 7.4. Mg^<2+> inhibited ATP formation. For efficient ATP production, it was necessary to maintain the concentration of soluble phosphate between 35 mM and 150 mM. A practical method fulfilling this requirement was developed, in which disodium phosphate was added to the reaction mixture intermittently every hour, starting at the third hour of the reaction. Under these optimum conditions, 13 mg of ATP Na_s・3H_2O per ml was produced from 3.5 mg of adenine per ml. The molar ratio of the transformation of adenine to ATP was approximately 83%.
