Purification and Characterization of β-Galactosidase from Scopulariopsis sp.

A strain of Scopulariopsis sp. which produced high activity of extracellular β-galactosidase was isolated from soil, and the crude enzyme was propared by wheat bran culture of the microorganism. The crude enzyme was further purified by precipitation with ammonium sulfate, and DEAE-cellulose, and CM-cellullose column chromatographies. The purified β-galactosidase was electrophoretically homogeneous and its molecular weight was estimated to be 95,000 by both Sephadex G-200 gel filtration and SDS-polyacrylamide gel electrophoresis. The optimum pH of the enzyme activity was between 3.6 and 5.0 and maximum activity appeared at 65℃.
