To evaluate the actual aeration efficiencies of different types of shaken flasks with various kinds of closures, both the volumetric oxygen transfer coefficients through the closure(K_1), and across the gas-liquid interface(K_2)were measured under shaking conditions by using a polarographic oxygen analyzer with membrane-type electrode. As a measure of aeration efficiency in shaken flask cultures, the aeration coefficient defined by[(1/K_1+1/K_2)^<-1.>1/V_l]was used. A wide range of aeration coefficients was obtained by various combinations of equipment and operating conditions. Also, shaken flask performance could be markedly improved by selecting appropriate conditions. The improvements of aeration efficiency were confirmed by an aerobic fermentation, i.e., the conversion of sorbitol to sorbose by Acetobacter suboxydans. The influence of aeration coefficient on the conversion rate was investigated, and the conversion rate was found to be stoichiometrically proportional to the overall oxygen transfer rate calculated from the aeration coefficient.
