Photocontrol of Trypsin-spiropyran Membrane Activity

Trypsin (EC 3.4.4.4) was immobilized in collagen membrane modified with β-1-[3,3-dimethyl-6'-nitrospiro(indoline-2,2'-2H-benzopyran)] propionic anhydride. The trypsin-spiropyran (collagen) membrane showed reverse photochromism. The specific activity of trypsin in the spiropyran membrane was 10% of that of native trypsin. The activity of the trypsin-spiropyran membrane under visible light was decreased to 78% of that in the dark. The optimum pH of the trypsin-spiropyran membrane was higher than that of native trypsin. The apparent Michaelis constant (K_m') of the trypsin-spiropyran membrane in the dark ws not changed with visible light irradiation. The apparant diffusion coefficient of the substrate in the dark was larger than that under visible light.
