For the optimum design of micro-fluidic devices, a well understanding of diffusive transport processes performs a critical contribution. Especially, the diffusion coefficient is the important property for the numerical simulation. In this article, we report the development of the measurement system for the diffusion coefficient in micro-scale, which combines the Soret effect with the diffraction or the heterodyne interferometry. By the Soret effect, solute molecules are simply driven by temperature gradient without flux of solvent. In order to detect the diffusion phenomenon with high spatial and time resolution, we have studied two observing techniques based on the diffraction or heterodyne interferometry. In the method using the diffraction, we have measured molar mass dependences of mass diffusion coefficient for high-density polymer solutions. Then we adopt the heterodyne interferometer with a stabilization technique for the measurement system. According to the experimental results, the effectiveness of differential detecting scheme is demonstrated, and the ability of the microscopic optical setup combined with the heterodyne interferometer is confirmed. We may conclude that the detecting technique has the possibility to measure the micro-scale measurement of the diffusion coefficient, and it is necessary to optimize the experimental parameters.
