Retrograde Perfusion of the Cerebral Vein with Antioxidant L Y231617 Reduces Brain Damage in the Rat Focal Ischemia Model

• 抗酸化剤LY231617のラットにおける中大脳動脈3時間虚血モデルに対する効果を逆行性脳静脈灌流(RPCV)による投与と全身投与で比較した。RPCVはラットの中大脳動脈領域からの血流を受けている下大脳静脈に逆行性にカニューレを挿入し薬剤を投与する方法で虚血3時間後より2時間の投与を行った。投与終了後オートラジオグラフィによる局所脳血流および、間接的にプロテインカイネースCの活性を現わしていると思われるフォルボールエステルの結合能、さらに虚血の大きさを定量的に判定した。RPCVによる本剤の投与は経静脈的全身投与に比べて、虚血巣内の血流量を増加し、フォルボールエステルの結合能を低下させており、有為に虚血範囲が縮小していた。
