MORPHOLOGICAL STUDIES ON THE ESTABLISHED CELL LINES OBTAINED FROM NORMAL HUMAN DECIDUAL TISSUE OF AN EARLY STAGE OF GESTATION

正常妊娠初期の脱落膜組織細胞を継代培養し, epithelial-like cell line [TTK-1 (E)] および fibroblast-like cell line [TTK-1 (F)] の二つの細胞系をえた. 初代培養は1979年7月に組織片培養法で開始し以後約2カ月ごとに継代培養を繰返したところ, epithelial-like cellが優勢なものとfibroblast-like cellが優勢なものとが出現し, 1年後にはそれぞれ1種類の細胞からなる培養がえられた. 前者は1988年4月現在まで9年間ほぼ一定の速度で活発な増殖を続け継代培養により維持されており, TTK-1 (E) cell lineと名付けた. 後者は次第に増殖力が低下し約3年後にほほとんど増殖を停止した. 継代培養を中止し培地交換のみを続けていたところ4カ月後に突然活発な細胞増殖が起こり, 以後今日まで活発な増殖を示し, 継代培養されており, TTK-1 (F) cell lineと名付けた. 位相差光顕, 走査電顕, 透過電顕による観察では, TTK-1 (E)は上皮細胞様, TTK-1 (F)は線維芽細胞様の所見を示し, 染色体検索では両者とも異数性で, ヌードマウスへの移植実験ではともに高い腫瘍形成率を示し未分化な細胞によりなる索状ないし腺癌様構造がみられた. この結果から両細胞系ほ継代培養中に自然発生したneoplastic transformationによつて樹立されたと考えられ, 子宮内膜由来の悪性腫蕩発生に関する細胞レベルでの解明に有用な実験モデルとなりうると思われる.
