A method of calculating mass splitting is formulated within the framework of the S-matrix theory. It is applied to calculation of the proton-neutron mass difference. In the approximation of taking the nucleon-antinucleon intermediate states, the result is much the same as that of Feynman-Speisman picture. In the approximation of taking account of mesonic resonant intermediate states, the predicted value is qualitatively in a good agreement with experiment. The same formulation is also applied to mass splitting of the baryon supermultiplets.
