[SHORT REPORTS]Negative Dromotropic Effects of SD-3212, a New Antiarrhythmic Agent, on Anisotropic Cardiac Muscle

The influence of myocardial fiber orientation on the inhibitory efects of SD3121, a novel Class-I antiarrhythmic agent possessing a high affinity to inactivated sodium channels, on conduction was investigated in isolated rabbit ventricular muscle. In the control, conduction velocity (θ) during longitudinal (L) propagation along the fiber orientation was 3.1-3.4 times higher than during transverse (T) propagation across the fibers. SD-3212 (3 μM and 10 μM) caused a concentration-dependent decrease of θ in both the L and T propagations at stimulation frequencies ≧ 1.0 Hz. The drug-induced θ reduction was similar for the L and T propagations, indicating that the anisotropic ratio of θ (θL/θT) is unaffected by the drug. These results in combination with our previous reports suggest a close relationship between the directional differences in the nagative dromotropic effects of antiarrhythmic drugs and the state-dependence of their sodium channel blocking actions.
